5-HTP is the direct precursor to serotonin in the brain. Unlike tryptophan, which requires an extra conversion step, 5-HTP crosses the blood-brain barrier efficiently and converts to serotonin with high reliability. A 2002 Cochrane review found that while the evidence for 5-HTP in depression is promising, the trials were generally small. It is not a pharmaceutical antidepressant and should not be treated as one. For adults dealing with stress-related mood fluctuations rather than clinical depression, the research is more consistently supportive.
On sleep: a 2009 study in the European Journal of Neuroscience found that 5-HTP supplementation increased REM sleep duration. The melatonin pathway is the likely mechanism — serotonin is the direct biochemical precursor to melatonin.
The appetite research is compelling. A double-blind, placebo-controlled trial published in the American Journal of Clinical Nutrition (1992) found that obese subjects taking 5-HTP before meals consumed significantly fewer calories and lost more weight over six weeks than placebo, without any dietary instruction.
Important safety note: Do not take 5-HTP alongside SSRIs, SNRIs, MAOIs, or other serotonergic medications. The combination can cause excess serotonin activity. Consult your doctor before use if you take any prescription medications.
Suggested use: One capsule (100mg) taken 30-45 minutes before bed. Start here and assess tolerance before increasing dose.
